Phosphoinositides: Functions in autophagy-related stress responses

چکیده
Phosphoinositides are key lipids in eukaryotes, regulating organelles' identity and function. Their synthesis turnover require specific phosphorylation/dephosphorylation events that ensured by dedicated lipid kinases phosphatases, which modulate the structure of inositol ring adding or removing phosphates on positions 3, 4 5. Beside their implication intracellular signalization cytoskeleton dynamics, phosphoinositides essential for vesicular transport along trafficking routes, providing molecular scaffolds to membrane related such as budding, fission fusion. Robust detailed literature demonstrated some members family crucial autophagy pathway, acting fine tuners regulators. In this review, we discuss known functions canonical processes, during autophagosome formation, well importance organelle-based processes directly connected autophagic machinery, endosomal ciliogenesis innate immunity.

Autophagosome formation is stimulated by canonical VPS34-dependent formation of phosphatidylinositol 3-phosphate [PI(3)P], which recruits effectors such as WIPI2. However, non-canonical VPS34-independent autophagy has also been proposed. We recently described that PI(5)P regulates autophagosome biogenesis, recruits WIPI2, and rescues autophagy in VPS34-inactivated cells. These alternative autop...

MicroRNAs (miRNAs) are a class of ∼22 nucleotide short noncoding RNAs that play key roles in fundamental cellular processes, including how cells respond to changes in environment or, broadly defined, stresses. Responding to stresses, cells either choose to restore or reprogram their gene expression patterns. This decision is partly mediated by miRNA functions, in particular by modulating the am...

Macroautophagy is a process in which cytoplasmic components, including whole organelles, are degraded within lysosomes. Basally, this process is essential for homeostasis and is constitutively functional in most cells, but it can also be implemented as part of stress responses. We discuss findings showing that autophagy proteins can modulate and amplify the activities of transcription factors i...
متن کاملRegulation of innate immune responses by autophagy-related proteins
Pattern recognition receptors detect microbial components and induce innate immune responses, the first line of host defense against infectious agents. However, aberrant activation of immune responses often causes massive inflammation, leading to the development of autoimmune diseases. Therefore, both activation and inactivation of innate immune responses must be strictly controlled. Recent stu...
متن کاملA Role for Phosphoinositides in Regulating Plant Nuclear Functions
Nuclear localized inositol phospholipids and inositol phosphates are important for regulating many essential processes in animal and yeast cells such as DNA replication, recombination, RNA processing, mRNA export and cell cycle progression. An overview of the current literature indicates the presence of a plant nuclear phosphoinositide (PI) pathway. Inositol phospholipids, inositol phosphates, ...
